Diatomaceous earth is an organic naturally occurring material rich in silicon. This silicon source can be used in organic agriculture, it also has a great potential of use in the acclimatization of crops. However, there are no reports of the effects of diatomaceous earth supplementation on the micropropagation of sweet potato. Thus, the objective of this study was to evaluate the effects of different concentrations of diatomaceous earth applied in vitro on the growth, physiology and anatomy of sweet potato cv. 'Brazlândia Branca' after acclimatization. Four concentrations of diatomaceous earth. After 30 days of in vitro growth, the plants were transferred to a greenhouse for acclimatization. After 45 days, leaf number, shoot and root length, fresh and dry shoot and root mass, gas exchange, chlorophyll content, root and leaf anatomy. The experimental design was completely randomized. The supplementation of diatomaceous earth in the in vitro cultivation had beneficial effects, increasing the accumulation of mass, improving the photosynthetic apparatus and promoting favorable anatomical characteristics during the acclimatization of the sweet potato plants. In addition, the use of diatomaceous earth achieved adequate seedling development, with higher seedling quality and resistance to biotic and abiotic effects than attained with control treatment.

Strawberry (Fragaria x ananassa, Duch.) is an important crop worldwide. However, since it is a highly demanding crop in terms of the chemical conditions of the substrate, a large part of strawberry production implies the application of large amounts of fertilizers in the production fields. This practice can cause environmental problems, in addition to increases in the fruit's production costs. In this context, applying plant growth-promoting bacteria in production fields can be an essential strategy, especially thanks to their ability to stimulate plant growth via different mechanisms. Therefore, this study aimed to test in vitro and in vivo the potential of bacteria isolated from strawberry leaves and roots to directly promote plant growth. The isolates were tested in vitro for their ability to produce auxins, solubilize phosphate and fix nitrogen. Isolates selected in vitro were tested on strawberry plants to promote plant growth and increase the accumulation of nitrogen and phosphorus in the leaves. The tested isolates showed an effect on plant growth according to biometric parameters. Among the tested isolates, more expressive results for the studied variables were observed with the inoculation of the isolate MET12M2, belonging to the species Brevibacillus fluminis. In general, bacterial inoculation induced strain-dependent effects on strawberry growth. In vitro and in vivo assays showed the potential use of the B. fluminis MET12M2 isolate as a growth promoter for strawberries.

To produce pitaya (Hylocereus spp.), it is essential that pollination occur, either artificially or by pollinating agents. This study evaluated the viability of self-pollination, cross-pollination, and natural pollination, as well as pollen viability, stigma receptivity and ideal pollination window in pitaya flowers (Hylocereus spp.). An experiment was conducted with a randomized block design consisting of four treatments: T1: manual self-pollination; T2: nocturnal open pollination; T3: diurnal open pollination; and T4: manual cross-pollination - red-fleshed pitaya [ Hylocereus polyrhizus] pollen placed on white-fleshed pitaya [ Hylocereus undatus] stigma. The experiment had four replicates, with two plants per replicate (two flowers per plant), totaling 16 flowers per treatment. The analyzed variables were fruit weight, length, diameter, titratable acidity, pH, soluble solids, and SS/TA ratio; the germination percentage of pollen grains; and the receptivity of stigmas. Artificial pollination (self- and cross-pollination) of white-fleshed pitaya (Hylocereus undatus) is viable, resulting in larger fruits than natural pollination, with cross-pollination being the most recommended. Throughout the floral opening period, the pitaya flower (Hylocereus spp.) has receptive stigma capable of receiving pollen. The flowers exhibit the highest pollen germination rate at 7 p.m, which is the ideal pollination window for pitaya.

One of the largest accidents with mine tailings happened in Brazil in 2015, with the rupture of the Fundão dam, and the physical characteristics of these tailings make it difficult to recover degraded areas. Hymenaea courbaril is a tree species native to Brazil that has low nutritional and water requirements, besides its capacity for survival in contaminated environments. In this study we hypothesized that inoculation with diazotrophs would improve the growth and physiology of H. courbaril in tailings, favoring the reforestation process aiming the recovery of the accident site. Every 20 days for 60 days, we investigated the morphophysiology of H. courbaril grown in iron mine tailings or soil, with the addition of nitrate (N-positive control), non-inoculation (negative control) or inoculation with native diazotrophic bacteria previously isolated from the tailings (UNIFENAS100-569; UNIFENAS100-654 and UNIFENAS100-638). We found that H. courbaril has survival capacity under mine tailings, with no growth alteration in the tailings, although there were signs of reduced ability for photoprotective responses. Inoculation with diazotrophic bacteria improved physiological aspects of H. courbaril and strain UNIFENAS100-638 was the most effective in favoring total growth of plants, net photosynthetic rate and root morphology under mine tailings. The survival capacity and growth of H. courbaril indicates the possibility of its use for reforestation in areas degraded by mine tailings. Further studies are necessary in field conditions and with a larger experimental period to more thoroughly understand H. courbaril tolerance.

Currently, southern Minas Gerais (MG) state is an important producer of different olive tree (Olea europaea L.) cultivars because, in this region, the plants can differentiate the buds to produce flowers and fruit. To stimulate the rooting of cuttings, the synthetic hormone indole-3-butyric acid (IBA) at a concentration of 3 g L−1 is used commercially. However, few studies have investigated arbuscular mycorrhizal fungi (AMF), isolated or combined with rhizobacteria, as a biotechnological tool to produce hormones that function in the rooting of olive tree cuttings. The aim of this study was to evaluate the capacity of different AMF species (Rhizophagus clarus, Gigaspora rosea, or Acaulospora scrobiculata), combined or not with IBA or rhizobacteria, to promote the rooting of three olive tree cuttings (Arbequina, Grappolo 541, and Maria da Fé) with potential for cultivation in this region. For this, three experiments were conducted at the Experimental Farm of EPAMIG in Maria da Fé (MG), and the rooting potential of the olive tree cuttings inoculated with I) AMF, II) AMF combined with increasing doses of IBA, and III) AMF combined with three isolates of rhizobacteria was evaluated. The inoculation of olive tree cuttings of cultivars Arbequina, Grappolo 541, and Maria da Fé with Rhizophagus clarus, Gigaspora rosea, or Acaulospora scrobiculata combined or not with IBA or rhizobacteria did not significantly promote rooting. Alternative forms of rooting olive tree cuttings are still a challenge, and further studies for standardizing methodologies and experimental conditions are required.

The objective of this work was to carry out the in vitro establishment of Echynochloa polystachya aiming at obtaining a micropropagation protocol for works involving the selection of superior genotypes and the cultivation of the species. E. polystachya stems were collected in the municipality of Manaus-AM. Explants were inoculated in test tubes containing Murashige and Skoog (MS) medium. Thirty days after in vitro establishment, the rate of sprouting and contamination were evaluated. Experiments were also carried out to assess the effects of sucrose and 6-benzylaminopurine (BAP) concentrations on the tillering rate of explants. It was found that during the successive subcultures there was a decrease in internodes and the consequent loss of vigor. There were responses in the multiplication rate at concentrations starting from 45 g L-1 sucrose. In addition, BAP and sucrose interfered the development and in vitro multiplication. Sucrose in conjunction with BAP was harmful and shortened internodes. The physiological state of the explants for the species under study was intrinsically linked to the concentrations of sucrose used for the culture medium and the concentrations of BAP. However, the sucrose and BAP concentrations suggested for in vitro cultivation of E. Polystachya must be adjusted during successive subcultures. Absence of contamination in the in vitro establishment occurred at concentrations 15, 30 and 60 g L-1 sucrose. The combination of 1.5 mg L-1 BAP and 30 g L-1 sucrose promoted greater induction of sprouts. In addition, the in vitro rooting of E. polystachya was 45%.

Garlic is a health promoter that has important bioactive compounds. The bioactive extraction is an important step in the analysis of constituents present in plant preparations. The purpose of this study is to optimize the extraction with the best proportion of solvents to obtain total phenolic compounds (TPC) and thiosulfinates (TS) from dried garlic powder, and evaluate the antioxidant activities of the optimized extracts. A statistical mixture simplex axial design was used to evaluate the effect of solvents (water, ethanol, and acetone), as well as mixtures of these solvents, after two ultrasound extraction cycles of 15 min. Results showed that solvent mixtures with a high portion of water and pure water were efficient for TPC and TS recovery through this extraction procedure. According to the regression model computed, the most significant solvent mixtures to obtain high TPC and TS recovery from dried garlic powder are, respectively, the binary mixture with 75% water and 25% acetone and pure water. These optimized extracts presented oxygen radical absorbance capacity. Pure water was better for total antioxidant capacity, and the binary mixture of water-acetone (75:25) was better for DPPH scavenging activity. These optimized extracts can be used for industrial and research applications.

Botrytis cinerea causes the gray mold disease in a wide range of plant hosts, especially in post-harvest periods. The control of this phytopathogen has been accomplished through the application of fungicides. However, this practice can cause environmental problems and increase fruit production costs. In addition, this fungus species has developed resistance to conventional synthetic fungicides. In this context, plant growth-promoting bacteria have shown potential for application in agricultural production because they are able to stimulate plant growth through different mechanisms, including the biological control of phytopathogens (indirect growth promotion mechanism). The aim of this work was to evaluate in vitro and in fruits the potential for indirect plant growth-promotion of bacteria isolated from strawberry leaves and roots. Dual plate method and inverted plate method were used to verify the ability of controlling in vitro the growth of Botrytis cinerea via the production of diffusible and volatile antifungal compounds, respectively. The effect of six bacterial isolates that showed greater potential for biological control in vitro was evaluated by scanning electron microscopy. Antifungal compounds produced by these bacterial isolates were identified by liquid chromatography coupled with mass spectrometry. Six bacterial strains were tested on strawberry pseudofruits. Five selected strains belong to the genus Bacillus and one to the genus Pantoea sp. Selected strains were able to inhibit more than 80 % of the mycelial growth of B. cinerea by the production of diffusible compounds and 90 % by volatile antifungal compounds production. Scanning electron microscopy showed the intense degradation of fungal hyphae caused by the presence of all bacterial strains. Bioactive compounds (salycilamide, maculosin, herniarin, lauroyl diethanolamide, baptifoline, undecanedioic acid, botrydial, 8 3-butylidene-7-hydroxyphthalide and N-(3-oxo-henoyl)-homoserine lactone) were obtained from liquid culture of these strains and extraction with ethyl acetate. All six isolates tested in vivo reduced the incidence of gray mold in strawberry pseudofruits in postharvest. It is concluded that isolates 26, 29, 65, 69, 132 (Bacillus sp.) and MQT16M1 (Pantoea sp.) have potential application for the biological control of Botrytis cinerea in strawberry via the production of diffusible and volatile antifungal compounds.

The association of different species of endophytic bacteria with the rhizosphere of the host plants can stimulate growth, development and acclimatization, offering a greater quantity of seedlings, in addition to reducing the cycle, providing economic return to the producer. The objective of this study was to evaluate the effect of introduction four bacterial isolates through inoculation into the root system in three banana cultivars (Prata Anã, Grande Naine and BRS Princesa) in the acclimatization phase. The evaluated treatments were: control (nutrient broth without bacteria); Bacillus cereus strain 1 (BC1); Bacillus cereus strain 2 (BC2); Bacillus thuringiensis (BT); Buttiauxella agrestis (BA). The morphological characteristics related to the development of the plants (total height and pseudostem diameter) were evaluated throughout the acclimatization period. After 90 days of transplanting and acclimatization, root length, leaf number, dry root weight, pseudostem and leaf, leaf area, internal carbon concentration, stomatal conductance, photosynthesis rate, transpiration rate, leaf temperature and chlorophyll were evaluated. The bacteria showed different results in relation to the studied cultivars. Considering the morphological and physiological characteristics observed in this study, B. thuringiensis for the cultivars Prata Anã and Grande Naine and the B. agrestis for the cultivar BRS Princesa are recommended for the process of acclimatization of banana seedlings, as they stimulated growth of the plant, increasing the dry mass, besides promoting the growth of roots. In this way, they improved the physiological aspects of the plants and reduced the period of acclimatization of the banana.

Although the demand for organic fertilizers in agriculture is growing, it remains incipient in some production sectors, such as medicinal plant production, which does not possess specific technology for its development. Solid residues are highly contaminant to water sources, soil, populations and biodiversity but can potentially be used to produce organic composts and vermicompost, such production not only enables nutrient recycling but also acts on soil conditioning, increasing soil organic matter and improving its physical, chemical and biological characteristics, as well as strengthening organic production with an emphasis on medicinal plant production. In this context, recommendations for and applications of organic fertilizers available in the market were reviewed; in addition, the imminent socioeconomic demand for organic compost and vermicompost production was contextualized based on residues from coffee and sugarcane production and cattle farming for application to the medicinal plant production chain. It is concluded that although these sectors produce a considerable amount of residues, they are not being reused in formulation of organic composts and vermicompost, and commercial organic fertilizers recommended for medicinal plant production were not found. Thus, the formulation of vermicomposting and composting from the reuse of agricultural residues, are potential social, economic technologies and tools to be valued and disseminated to traditional and family farmers. Embora a demanda por fertilizantes orgânicos na agricultura seja crescente, ela ainda é incipiente em alguns setores produtivos, como a produção de plantas medicinais, que não possui tecnologia específica para seu desenvolvimento. Os resíduos sólidos são altamente contaminantes para fontes de água, solo, populações e biodiversidade, mas podem ser potencialmente utilizados para a produção de compostos orgânicos e vermicomposto, tal produção não só possibilita a reciclagem de nutrientes, mas também atua no condicionamento do solo, aumentando a matéria orgânica do solo e melhorando seu aspecto físico, químico e características biológicas, além do fortalecimento da produção orgânica com ênfase na produção de plantas medicinais. Nesse contexto, foram revisadas as recomendações e aplicações dos fertilizantes orgânicos disponíveis no mercado; além disso, a iminente demanda socioeconômica pela produção de composto orgânico e vermicomposto foi contextualizada a partir dos resíduos da produção de café, cana-de-açúcar e pecuária para aplicação na cadeia produtiva de plantas medicinais. Conclui-se que embora esses setores produzam uma quantidade considerável de resíduos, eles não estão sendo reaproveitados na formulação de compostos orgânicos e vermicomposto, e não foram encontrados fertilizantes orgânicos comerciais recomendados para a produção de plantas medicinais. Sendo assim, a formulação de vermicompostagem e compostagem a partir do reaproveitamento de resíduos agropecuários, são potenciais tecnologias sociais, econômicas e ferramentas a serem valorizadas e disseminadas para os agricultores tradicionais e familiares. Garlic (Allium sativum L.) is one of the crops whose economic importance has increased considerably in recent years in Brazil. The use of plant growth-promoting bacteria (PGPB) is a useful alternative for reducing the cost of agricultural inputs and, consequently, for increasing productivity. Therefore, the main objective of this work was to isolate and evaluate potential growth promoters in plants and plant pathogenic fungi growth inhibitors using endophytic bacteria from garlic roots and bacteria from the Agricultural Microbiology Culture Collection at the Federal University of Lavras. Besides verifying improvements in the growth and physiology of garlic meristems grown in vitro under the action of PGPB. Forty-eight endophytic bacteria were identified using matrix-assisted laser desorption/ionization-time of flight mass spectrometry based on the protein profile of each isolate. Four isolates were chosen according to their ability to fix nitrogen, to produce auxin and solubilize phosphate. The cultivation of garlic meristems in tissue culture with these bacteria was established at a population level of 106 CFU/mL. The evaluated criteria were: (1) the colonization capacity of the bacteria inside the garlic plants determined through scanning electron microscopy; (2) the chlorophyll content; and (3) the growth of garlic plants in vitro post-PGPB inoculation. Volatiles emitted by those isolates inhibited fungi growth. The inoculation of garlic meristems with Enterobacter cloacae and Burkholderia cepacia promoted an improvement in the growth and physiological attributes of garlic, indicating the feasibility of their application as plant growth promoters for commercial cultivation.

The collapse of the Fundão dam in Mariana, MG, in 2015 resulted in the overflow of more than 50 million m3 of mud containing mine tailings, leaving a path of destruction and immeasurable social and environmental consequences. Tailings' chemical and physical assessments revealed the presence of some elements at levels higher than those allowed by Brazilian guidelines. The tailings also showed high density, which restricts vegetation recovery. Therefore, this study aimed to analyze the effects of mud containing mine tailings from the Fundão dam on the germination and initial growth and development of three plant species: millet, maize, and sorghum. These species were cultivated on substrates with five tailings proportions: 0 T (100% sand), 25 T (25% tailings + 75% sand); 50 T (50% tailings + 50% sand); 75 T (75% tailings + 25% sand); and 100 T (100% tailings). In experiment I, the germination and initial growth of seedlings (plants with 1 or none fully expanded leaf) in these substrates were evaluated. In experiment II, growth parameters, photosynthetic efficiency (gas exchange and chlorophyll a fluorescence), metal accumulation, and plant root morphology of the same species were evaluated at the three fully expanded leaves vegetative stage (V3). Overall, the germination of seedlings and the initial growth of the three species analyzed were not affected by the presence of tailings. However, in plants at the V3 stage, morphophysiology variations differed among species, given that their growth, biomass accumulation, and root dynamics were altered. Proportions of tailings in the substrate did not influence the absorption of iron or manganese by the studied plants. At the V3 stage, maize was the most tolerant, with a more robust root system, and showed fewer morphological changes and greater water use efficiency than the other studied species.

Morphological and agronomical describers are traditionally used in plant characterization. However, the usage of these describers have some limitations such as susceptibility to abiotic and biotic stress and environmental factors. Furthermore, the describers are not stable over time and many can only be evaluated during the adult phase of the plants, which requires time and physical space. Molecular markers offer numerous advantages compared to the conventional alternatives based on phenotype: they are stable and detectable in all vegetable tissues, and are independent of the environment and development phase. One of the main advantages of the use of molecular markers is the time reduction in the identification of genetic diversity among the studied subjects, as the genotypes may even be described for the seed or seedling phase. Many countries have already adopted molecular markers to identify olive cultivars more accurately. The aim of this study was to evaluate the genetic identity of eight olive accessions supposedly belonging to cultivar Arbequina by using microsatellite (SSR) and Sequence Characterized Amplified Region (SCAR) markers. One accession corresponding to the cultivar was also incorporated into the analysis as a reference genotype. The molecular marker data were analyzed on the software GENALEX6.The markers generated an accumulated PI and PE of 1.26 x 10-6 and 0.949, respectively.The results supported the hypothesis that all accessions belong to the cultivar Arbequina, and the markers can therefore be applied to other varieties of olive species.


Descritores morfológicos e agronômicos são tradicionalmente utilizados na caracterização de plantas. Apesar de recomendado, o emprego destes descritores apresenta algumas limitações como a influência a estresses abióticos e bióticos e aos efeitos do ambiente. Além disso, não são estáveis ao longo do tempo e muitos só podem ser avaliados durante a fase adulta das plantas, o que requer tempo e espaço físico para as avaliações. Os marcadores moleculares oferecem numerosas vantagens relativamente às alternativas convencionais baseadas no fenótipo, pois são estáveis e detectáveis em todos os tecidos vegetais, independente do ambiente e fase de desenvolvimento e uma das principais vantagens da utilização destes é proporcionar a redução do tempo na identificação da diversidade genética entre os indivíduos trabalhados, podendo ser avaliadas genótipos ainda na fase de semente ou de plântula. O objetivo deste estudo foi avaliar a identidade genética de oito acessos de oliveira supostamente pertencentes a cultivar Arbequina usando microssatélites (SSR) e Sequence Characterized Amplified Region (SCAR) marcadores. Um acesso correspondente a cultivar também foi incorporado na análise como o genótipo de referência. Os dados de marcadores moleculares foram analisados com o software GENALEX 6. Como resultado, os marcadores SSR geraram um PI acumulada e PE de 1,26 x 10- 6 e 0,949, respectivamente. Os resultados suportam a hipótese de que todos os acessos pertencem a cultivar Arbequina, e, por conseguinte, esses marcadores podem ser aplicados em situações semelhantes em outras variedades de espécies de oliveira.

The biotechnological interest in genus Physalis has increased in the last decades, however, there are still few micropropagation studies of this genus. The objective of this study was to evaluate P. angulata photoautotrophic and photomixotrophic micropropagation with gas exchange under seven light spectra and five concentrations of sucrose. Lighting were yellow, blue, white, red, green, red + blue LEDs and natural light filtered by mesh. Sucrose concentrations were 0, 7.5, 15, 22.5 and 30 g.L-1. Phytotechnical, anatomical features and photopigment contents were evaluated through stem and root segment length, leaf number, leaf area, chlorophyll a and b contents, carotenoids, adaxial epidermis, palisadic and spongy parenchyma and abaxial epidermis. The data were compared by Scott-Knott's mean test and principal components analysis using the R software. Comparing the variables within lighting types, it was observed that only the screen treatment, screen-filtered natural illumination, obtained assessment in all variables. Comparing the levels of sucrose, it was observed that the treatment 15 g.L-1 sucrose obtained the highest number of averages with maximum evaluation. It was concluded that the natural light filtered by screen with 50% of shading allowed the photoautotrophic micropropagation of P. angulata. Better development results were observed in photomixotrophic micropropagation with 15 g.L-1 of sucrose.


O interesse biotecnológico em Physalis aumentou nas últimas décadas, porém, ainda existem poucos trabalhos de micropropagação desse gênero. Objetivou-se avaliar sua micropropagação fotoautotrófica e fotomixotrófica com troca gasosa sob sete tipos de iluminação e cinco concentrações de sacarose. Foram utilizados LEDs amarelo, azul, branco, vermelho, verde, vermelho + azul e luz natural filtrada por malha. As concentrações de sacarose foram 0, 7,5, 15, 22,5 e 30 g.L-1. Características fitotécnicas, anatômicas e teor de fotopigmentos foram avaliados através de comprimento de segmento de caule e raíz, número de folhas, área foliar, teores de clorofilas a e b, carotenoides, epiderme adaxial, parênquimas paliçádico e esponjoso e epiderme abaxial. Os dados foram comparados por teste de média Scott-Knott e análise de componentes principais utilizando-se o software R. Comparando-se as variáveis dentro de tipos de iluminação, observou-se que apenas o tratamento screen, iluminação natural filtrada por malha, obteve avaliação máxima em todas as variáveis. Comparando-se os níveis de sucrose, observou-se que o tratamento 15 g.L-1 sacarose obteve o maior número de médias com avaliação máxima. Concluiu-se que a luz natural filtrada por tela com 50% de sombreamento permitiu a micropropagação fotoautotrófica de P. angulata. Observou-se melhores resultados de desenvolvimento na micropropagação fotomixotrófica com 15 g.L-1 de sacarose.

Physalis alkekengi is an ornamental plant that can also be used as a medicinal plant due to its anti-inflammatory, bactericidal, antitumor and fungicidal properties. Polyploidization can be an important tool in the genetic improvement of this species. The objective this work was to obtain tetraploids in vitro and to evaluate the phytotechnical traits of P. alkekengi. For this, nodal segments of P. alkekengi var. Franchettii were inoculated into petri dishes containing 100 ml of MS medium supplemented with colchicine at concentrations 0; 0.04; 0.08; 0.12; and 0.16% and kept in the dark for 24 and 48h. After the respective treatment periods with colchicine the segments were inoculated into test tubes. The tetraploids were identified by flow cytometry and classical cytogenetics. In vitro seedlings were measured: root length, nodal segment length, leaflet number and total leaf area. In the acclimatization phase, the area of the second leaf and total leaf, petiole radius, stem length, fruit weight with calyx, without calyx, fruit diameter, number of seeds and brix of the pulp were evaluated. Chlorophyll a, chlorophyll b, total chlorophyll, total carotenoid, total chlorophyll / total carotenoid ratio and chlorophyll a / b ratio were also estimated. The treatment that most produced tetraploid seedlings was with 0.08% colchicine per 24h. No significant difference was observed in 7 (seven) variables, these being all variables of photopigments, stem diameter (steam) and brix. In general, diploid (2x) plants were better in 9 (nine) while tetraploid seedlings were better in 6 (six) of the phytotechnical variables. It was concluded that the MS medium supplemented with 0.08% colchicine for 24 h allowed P. alkekengi tetraploides to be obtained with better phytotechnical qualities.


Physalis alkekengi é uma planta ornamental que também pode ser usada como planta medicinal devido às suas propriedades anti-inflamatórias, bactericidas, antitumorais e fungicidas. A poliploidização pode ser uma ferramenta importante para o melhoramento genético dessa espécie. O objetivo deste trabalho foi obter tetraplóides in vitro e avaliar as características fitotécnicas de P. alkekengi. Para isso, segmentos nodais de P. alkekengi var. Franchettii foram inoculados em placas de Petri contendo 100 ml de meio MS suplementado com colchicina nas concentrações 0; 0,04; 0,08; 0,12; e 0,16% e mantido no escuro por 24 e 48h. Após os respectivos períodos de tratamento com colchicina, os segmentos foram inoculados em tubos de ensaio. Os tetraplóides foram identificados por citometria de fluxo e citogenética clássica. As plântulas in vitro foram medidas: comprimento da raiz, comprimento do segmento nodal, número de folhetos e área foliar total. Na fase de aclimatação foram avaliadas a área da segunda folha e área foliar total, raio do pecíolo, comprimento do caule, peso do fruto com cálice, sem cálice, diâmetro do fruto, número de sementes e brix da polpa. Também foram estimadas clorofila a, clorofila b, clorofila total, carotenóides totais, razão clorofila total / carotenóide total e razão clorofila a / b. O tratamento que mais produziu mudas tetraplóides foi com colchicina a 0,08% por 24 horas. Não foi observada diferença significativa em 7 (sete) variáveis, sendo todas variáveis de fotopigmentos, diâmetro do caule (vapor) e brix. Em geral, as plantas diplóides (2x) foram melhores em 9 (nove) variáveis fitotécnicas, enquanto as mudas tetraplóides foram melhores em 6 (seis). Concluiu-se que o meio MS suplementado com colchicina a 0,08% por 24 h permitiu obter tetraploides de P. alkekengi com melhores qualidades fitotécnicas.

The Fundão dam was designed to store iron mine tailings in the region of Mariana, MG, Brazil. When it ruptured, the tailings overflowed. These tailings affected the soil due to the formation of a thick crust as a result of drying (compaction) and hindered the natural revegetation process. In this context, the use of organic fertilizers, including vermicompost, is method of reducing the physical limitations on root growth caused by soil properties and changing soil-metal interactions. For this reason, vermicompost was added to iron mine tailings, and its morphological and physiological effects on maize, millet and sorghum plants were studied. The experiment was conducted in a greenhouse using 6 dm3 pots. The plants were subjected to three treatments: mine tailings, mine tailings + vermicompost, and a reference soil. From the V3 stage onwards, biweekly growth, leaf gas exchange and chlorophyll fluorescence evaluations were performed. At the end of the experiment, dry biomass and metal, macro- and micronutrient contents were quantified, and the root morphology was evaluated. The tailings created physical limitations on root growth and had low nutrient content as well as high concentrations of chromium, iron and manganese. The addition of vermicompost favored increases in shoot and root dry biomass, increases in root length, volume, surface area and diameter, and the absorption of macro- and micronutrients, which was reflected in the growth of the studied species. In addition, vermicompost led to greater investment in thick and very thick roots, and in general, the plants showed no symptoms of metal toxicity. Considering the characteristics of the studied tailings, it can be concluded that vermicompost favors the growth of plant species and may be a viable method for beginning the recovery process in areas containing iron mine tailings.

Limitation to root growth results from forces required to overcome soil resistance to deformation. The variations in individual particle forces affects root development and often deflects the growth trajectory. We have developed transparent soil and optical projection tomography microscopy systems where measurements of growth trajectory and particle forces can be acquired in a granular medium at a range of confining pressures. We developed image-processing pipelines to analyse patterns in root trajectories and a stochastic-mechanical theory to establish how root deflections relate to particle forces and thickening of the root. Root thickening compensates for the increase in mean particle forces but does not prevent deflections from 5% of most extreme individual particle forces causing root deflection. The magnitude of deflections increases with pressure but they assemble into helices of conserved wavelength in response linked to gravitropism. The study reveals mechanisms for the understanding of root growth in mechanically impeding soil conditions and provides insights relevant to breeding of drought-resistant crops.

Plant growth-promoting bacteria have been highlighted by their potential for application in plant production, allowing the reduction of the use of fertilizers and pesticides, which is due to the ability to stimulate the growth of plants by nitrogen-fixation and production of phytohormones, such as indole-3-acetic acid (IAA). The objective of this study was to verify the potential of plant growth promotion of 25 wild isolates from the Agricultural Microbiology Culture Collection of the Federal University of Lavras (CCMA-UFLA) through the evaluation of the biological nitrogen-fixation capacity and the production of IAA. In addition, the growth of three selected strains inoculated on roots of strawberry seedlings in greenhouse conditions was evaluated. The experiment was conducted in a completely randomized design (CRD), with an 8 × 2 factorial schemes involving eight combinations of bacteria: alone, in pairs and threes, plus the control without inoculation. Two fertilizer levels were used (0% and 50% of nitrogen), totaling 16 treatments with eight replicates each. After 75 days, variables such as root length, root dry weight, aerial part length, aerial part dry weight, leaf number, total dry mass and ultrastructural analysis of the inoculated and uninoculated roots, were evaluated. The results showed that the strawberry crop responded positively to inoculation with the three bacteria combined Azospirillum brasilense (Ab-V5) + Burkholderia cepacia (CCMA 0056) + Enterobacter cloacae (CCMA 1285) compared to the uninoculated controls. More expressive responses in terms of plant growth were observed in relation to the combined inoculation of the three bacterial strains plus fertilizer application with 50% of nitrogen.

Due to the importance of studying alternatives to reduce pepper post-harvest loss and verify the processing potential of the several cultivars/varieties available, the objective of this study was to characterize common peppers in Brazil, and to evaluate their potential for further processing into jelly form. More specifically, the most-consumed species and varieties in Brazil were investigated, that is: Habanero, Cheiro do Norte, Biquinho, Malagueta, Cayenne, Paprika and Dedo de Moça. Additionally, the drivers of liking for pepper jelly were sought so as to understand the desirable characteristics of this product. The different peppers were initially assessed by the following physical and physicochemical analyses: length, diameter, unit mass, total soluble solids, total titratable acidity, pH, color, total phenolic content, antioxidant activity, and vitamin C levels. Jellies made with different pepper types were also analyzed for total titratable acidity, pH, color, and texture profile, and a sensory study was conducted to elucidate the drivers of liking. As expected, different pepper types varied in physical and physicochemical properties, and the resulting jellies also differed in terms of physical, physicochemical, textural and sensory characteristics. In general, less pungent peppers (Biquinho, Paprika and Cheiro do Norte) were the most suitable for processing to produce jellies with more desirable attributes for consumers: reddish color, characteristic flavor and aroma of a pepper, sweet taste, and low pungency.

Garlic (Allium sativum L.) plays an important role in popular culture due to its dietary and medicinal uses. It is also used to produce a wide range of pharmacologically interesting molecules. Several pathogens affect garlic plants, especially Athelia (Sclerotium) rolfsii, a fungus that is widespread and causes large economic losses. It causes direct damage to crops and leads to plant stress, which induces secondary metabolite production in plants. The use of microorganisms as biocontrol agents may induce the production of beneficial metabolites in plants that will protect it and promote resistance to pathogen attack. In addition to suppressing disease, biological control agents may have elicitor effects that could induce an increase in the production of useful bioactive secondary metabolites in plants, some of which may be of pharmacological interest. Therefore, the search for new biological control agents should also consider their potential as elicitor agents. This paper presents an analysis of the biological control of Athelia (Sclerotium) rolfsii by antagonistic microrganisms, the potential of yeasts and bacteria of the genus Bacillus for the biocontrol of phytopathogens, microrganisms influence in nutritional and bioactive compounds content of interest to the pharmaceutical industry.
